Introduction: The analysis of DNA from ancient remains could be useful for archeological purposes, and their results use to be of great importance on the field. However, the application of these techniques implies the implementation of several criteria that ensure the result authenticity. Aim: The aim of this review is to determine some basic criteria for the excavation process and posterior manipulation for samples susceptible to be use on ancient DNA projects. Content: This publication reviews the basic concepts on DNA structure, the PCR technique, the principal advances of ancient DNA field and their most important limitations. Conclusions: The obtaining of authenticated results on ancient DNA studies depends on the application of certain criteria on the previous archaeological and bioanthropological work. Without these criteria, conclusions obtained from the ancient DNA results lose credibility in the scientific community. Mainly, it is important to avoid direct manipulation of the samples during excavation and subsequent analysis, and the establishment of proper storage conditions to avoid DNA degradation.
